Creation: Appeared in South Carolina in 1930, the same year it appeared nationally. US 321 entered S.C. at Bowling Green and fed south through York and Lowrys more or less as today, before veering east following the railroad tracks, which entered Chester on the northeast side of town. US 321 ended in Chester at US 21 (current SC 72-121 Business). This routing was a replacement for original SC 16.
Adjustments: In 1949, US 321 was rerouted at Lowrys to process due south to Chester. This had been SC 325 prior to 1948. The old way from Lowrys to Chester became S-12-1, although some of it is now part of SC 909.
In 1950, US 321 was substantially extended south, all the way to Hardeeville. This replaced: US 21 from Chester to Winnsboro; SC 218 from Winnsboro to Columbia; multiplex with US 21 from northern Columbia to Dixiana; replaced SC 5 from Dixiana all the way to Hardeeville.
Originally, US 321 in Columbia followed Main St to Elmwood, then west to Assembly, south to Gervais, and followed US 1 west across the Saluda River (this was a 6-plex of routes: US 1-21-176-321-378 and SC 215). US 321 followed US 1 to Charleston Hwy (SC 602's beginning) before heading south towards Dixiana.
US 321 was placed on a new bypass west of Winnsboro in 1951 or 1952, leaving behind US 321 Business.
Between 1954-58, US 321 was rerouted in Columbia to continue south on Assembly to Blossom St before heading west across the Saluda River on the current US 321 bridge. The old route remained US 1-378 and SC 602, with a piece of Charleston Hwy becoming a US 1-21 connector.
US 321 was placed on a new bypass west of York in 1957 or 1958, leaving behind a second US 321 Business.
US 321 was added to the Chester Bypass with SC 9, plus a little new construction northwest of town between 1962-64, leaving behind a third US 321 Business.
Also between 1962-64, US 321 was placed on new alignment to bypass Blackstock, leaving behind S-20-130 and S-12-130 (a rare instance of county numbers staying the same across a boundary)
US 321 was extended south to I-95 as a multiplex with US 17 about 1974.
US 321 was removed from Assembly St in Columbia and rerouted along Huger St like today in 1978 or 1979. Assembly St became an extended SC 48.
Improvements: Paved only in York County upon designation. The remainder of US 321 was paved in 1937 or 1938
The first multilane section of US 321 was in place iin 1950, with its US 1 multiplex.
The next section to be upgraded was from Columbia down to Dixiana (US 21-176-321 split)
By 1964, US 321 was 4-lane from Clover to the N.C. Line; Chester to the SC 97 SB split; most of southern Chester County; and Dixiana through Columbia to the US 21-321 north split
By 1967, US 321 was 4-laned from the US 21 NB split to I-20 and from York north to the SC 161 split.
Between 1968-70, US 321 was 4-laned from Woodward down to near Winnsboro and on the US 301 multiplex near Ulmer.
In 1971 or 1972, most of the Winnsboro Bypass was 4-laned.
Between 1974-77, US 321 was 4-laned from Gaston down to Swansea and on the SC-9 portion of the Chester Bypass
Between 1978-79, US 321 was 4-laned a couple miles north of I-20.
Between 1989-90, US 321 was 4-laned from Clover to SC 161 and from I-95 to SC 46.
Comment: US 321 essentially slices South Carolina in half.
US 321 could be a viable alternative to go from Columbia to Savannah.
