What ELISPOT assays tell you
ELISPOT assays
are used to detect very small amounts of cytokines secreted by T
lymphocytes in culture. It requires concentrating the cytokine
molecules in small areas of a culture well by having them bind to
anti-cytokine antibodies coating the plate. The cells are then
removed, after which an enzyme-labeled anti-cytokine and a chromogenic
substrate for the enzyme are used to graphically detect the bound
cytokines.
Through this technique,
one can count the number of cytokine-secreting cells in each well.
