Illinois 127 is a significant north-south route that connects many rural areas together, from the southernmost reaches of Illinois in Alexander County north to near Raymond, in Montgomery County.

Intersections from north to south:

IL127 begins at I-55,on a stretch where it is multiplexed with IL48

Then IL48 splits off to head northeast at Raymond

It joins IL16 east

Then it leaves IL16 to go south at Hillsboro

IL185 by Taylor Springs

IL127 joins IL140 east

Then it leaves south at Greenville

US40

I-70

IL143

US50 joins it south

Then US50 leaves to the east

IL161

IL177

I-64

IL15 at Nashville

Then IL127 intersects IL154 in Pinckneyville and IL13 joins from the west

Then those 2 routes together intersect IL152 while multiplexed

IL4

Then IL149 joins the 2 routes, forming a "tripleplex" near Murphysboro IL13 leaves to the east as IL127 and IL149 go west

IL127 leaves IL149 and goes south again at Murphysboro

IL127 joins IL146 east

IL127 leaves south again at Jonesboro

Finally, it ends at IL3.

It passes through these towns: Raymond, Butler, Hillsboro, Taylor Springs, Donnellson, Greenville, Carlyle, New Minden, Nashville, Pinckneyville, Vergennes, Murphysboro, Jonesboro, Mill Creek, Tamms, Unity.
