The main character of Rurouni Kenshin, Himura Kenshin doesn't look like a skilled swordsman.He's often much shorter than his opponents, and is often underestimated by his opponents . Kenshin looks younger than he really is,his real age 28 years, and he's very kind to others. However, once he becomes angry and starts fighting seriously, Kenshin's eyes change, and he gives off an aura which can literally stop an opponent cold. By useing powerful fighting style of Hiten Mitsurugi Ryu, Kenshin is almost undefeatable. However, Kenshin has devoted his life to helping and protecting people with a reversal blade, sword to atone for his sins in his past, and hasn't killed anyone since the time he took his vow.

Kamiya Kaoru is 18 years old, and is sometimes called the "tanuki" or "raccoon" girl. Kaoru owns a dojo and is an instructor at her school, both of which was left behind by her deceased father. She is also an above average swordswoman, and she uses her Kamiya Kasshin Ryu to protect people using a bokken, or wooden sword. She is also stubborn, easily gets jealous, and can cook food which the Kenshin-gumi describes as "barely edible". However, Kaoru is also very kind and chipper, and has fallen in love with Kenshin.

Yahiko is a ten year old boy samurai family. After being rescued by Kaoru and Kenshin from some thieves that were using him to steal money,After that Yahiko has lived at the Kamiya dojo to practices Kamiya Kasshin Style. Yahiko is skilled for his age and even managed to copy one of Kenshin's moves. Yahiko admires Kenshin very much and remains faithful to him, always willing to risk his life for Kenshin. Yahiko is also always there to defend Kenshin when anyone tries to insult him while hoping to grow up to be just like Kenshin.

Sagara Sanosuke is both Kenshin's friend and fighting partner. He is only 19 year old and is able to surpass many of his enemies with his brute strength, endurance, and speed. Sano was once an enemy of Kenshin and underestimated him because Kenshin looked weak to him. When they fought, he was surprised when Kenshin sliced his zanbantou sword in half and later defeated him. Since then, Sano has stuck by Kenshin's side using his fists and Futae no Kiwami. However, Sano isn't always faithful to the Kenshin-gumi since he always chooses his past life with the Sekihoutai over the present. He even took the lastname of his old boss, Sagara, and continues to wear "Aku" or "evil" on the back of his shirt.

Takani Megumi is the beautiful, young doctor at the age of 22.She was rescued by Kenshin and his companions. Megumi is the daughter of a famous doctor and has longed to follow in his footsteps. However, she was forced to make deadly opium and later tried to kill both the person forcing her and herself. Sano stopped her, and Kenshin told her to become the doctor she had always wanted to be so she could save lives to atone for those who had died from the opium. Since then, she has been an excellent, kind doctor far skilled for that time period and a good cook. However, she loves teasing and making Kaoru jealous by flirting with "Ken-san", and she enjoys making fun of Sano.
