Illinois 96 is a north-south route in extreme western Illinois, following closely by the Mississippi River for much of its length. In fact, the river can be seen from the road in parts. It begins in southern Henderson County and runs south to Kampsville, in Calhoun County.

IL9 joins from the east at Dallas City
IL9 leaves west to cross into Iowa at Niota
IL96 joins US136 east near Keokuk, Iowa and Hamilton, Illinois
IL96 leaves to head south again at Hamilton
IL61 at Ursa
IL96 joins US24 west for a short distance north of Quincy
Then IL96 splits off of US24
IL104 at Quincy
IL96 joins IL106 east at Hull
IL96 breaks off of IL106 at Kinderhook
US54 at Atlas
IL96 ends in the south at IL100 in Kampsville.
It passes through these towns: Lomax, Dallas City, Pontoosuc, Niota, Nauvoo, Hamilton, Lima, Marcelline, Ursa, Quincy, Payson, Plainville, Hull, Kinderhook, New Canton, Rockport, Pleasant Hill, and Kampsville.
